When scanning files for viruses you should do nothing else. Do not search for any on the Internet or use your e-mail system.

Files
Once you have updated your virus definitions go to where you saved your file, hover your mouse over the file or folder and right-click, select "Scan for Viruses" or just "Scan" allow that to complete.
Hint: you should created a folder called My Downloads in the folder called My Documents so that any file that comes in off the Internet goes into this folder. Your anti-virus and spy-ware removal tools can be set easier to monitor these files; instead of having to look through the entire computer.

If it passes with no viruses or spy-ware detected you can then open, listen, play or install the file. If it does have a virus you should immediately delete the file or folder and take note where you went and do not trust that person, or site that you downloaded it from.
